目的研究穿孔素是否参与了特发性血小板减少症(ITP)的发病机制。方法采用RT-PCR法检测了35例特发性血小板减少症患者和45例健康个体外周血单个核细胞(PBMC)内穿孔素mRNA的表达,以免疫印迹法检测穿孔素蛋白含量。采用ELISA法检测受试对象血浆IL-4、IL-10、IFN-γ、TNF-α水平。以Spearman法分析穿孔素与上述细胞因子以及血小板计数的关系。结果与健康对照组比较,ITP患者PBMC中穿孔素蛋白和mRNA的表达均较健康个体增高(P〈0.01)。穿孔素蛋白水平与IFN-γ、TNF-α水平均呈正相关(P〈0.01),与血小板计数呈负相关(P〈0.05),与IL-4、IL-10水平无相关性(P〉0.05)。结论穿孔素参与了ITP的发病机制。
Objective To investigate whether perforin was involved in the pathogenesis of idiopathic thrombocytopenic purpura(ITP).Methods Perforin relative expressions in peripheral blood mononuclear cells(PBMCs) of 35 ITP patients and 45 healthy controls were detected by RT-PCR and Western blot.The relationship between perforin protein and platelet count,serum interleukin 4(IL-4),IL-10,tumor necrosis factor alpha(TNF-α) and interferon gamma(IFN-γ) were analyzed by spearman approach.Results Increased perforin,both at mRNA and protein level,was observed in ITP patients(P0.01).Perforin protein level in PBMCs was positively correlated with serum IFN-γ and TNF-α and negatively correlated with platelet count(P0.05).The correlations between perforin and IL-10,IL-4 did not achieve statistical significance(P0.05).Conclusion Perforin was involved in the pathogenesis of ITP.
