目的:建立兔眼翼状胬肉模型。方法:切除颞上象限LSCs+1.25%盐酸点眼刺激以建立兔眼翼状胬肉模型,每周观察角膜上皮缺损、结膜充血、新生组织生长情况。于第14周末取LSCs缺损的角膜缘标本(实验组)及同眼对侧角膜缘标本(对照组),共8个标本,行HE染色分析。结果:兔眼翼状胬肉模型未造成功,观察14周未见角膜缘新生组织,仅2眼穹窿结膜处少许增生。HE染色示:实验组角膜缘处上皮不平整、细胞大量增生,基底层组织疏松,有炎细胞;而对照组角膜缘结构几乎完整。结论:翼状胬肉模型失败有两种解释:首先翼状胬肉发病机制复杂,仅切除角膜缘+慢性盐酸刺激可能不足以导致翼状胬肉的发生;其次切除角膜缘+慢性盐酸刺激可以导致翼状胬肉的发生,而本实验所得结果为假阴性。翼状胬肉动物模型尚不成熟,待进一步探索研究。
Objective: To set up animal model of pterygium in rabbit eye. Methods: Removal of corneal limbal stem cells of superior temporal quadrant and stimulating of 1.25 percent diluted hydrochloric acid to establish pterygium model in rabbit eyes. The condition of corneal epithelial defect、conjunctival congestion and new organizations was observed every weekend. Rabbits were executed by end of the fourteenth week, and samples of corneal marginal were took respectively (four experimental group and four control group) for hematoxylin and eosin staining. Results: We didn’t build a successful pterygium model in rabbit eyes. There was no new limbal organization and only slightness proliferation of fornix conjunctive in two eyes within fourteen weeks. The result of HE staining showed a large number of hyperplasia of limbal epithelial cells, and the tissue of basal layer was loose and had inflammatory cells in the experimental group, while the structure of corneal limbus was almost integrity in the control group. Conclusion: There may be two possible explanations for the failure of its model: firstly: The pathogenesis of pterygium might be so complex that only by removal of corneal limbal stem cells and stimulation of hydrochloric acid can not lead to the occurrence of pterygium; secondly, pterygium may be leaded by removal of corneal limbal stem cells and stimulation of hydrochloric acid, and the result of our experiment are false-negative. The animal model of pterygium is not yet mature so far and need further study.
