目的观察中药糖肾清1号对糖尿病肾病肾组织局部纤维化因子表达的影响。方法 建立糖尿病肾病大鼠模型,设正常对照组、模型组、阳性对照药组、糖肾清1号甲、乙、丙、丁组及各组又分大、中、小剂量治疗组,分别采用RT-PCR、Western blot检测大鼠TGFβ1、CTGF mRNA转录及蛋白表达水平进行检测,采用光镜、电镜进行组织形态学检测。结果糖肾清1号呈量效依赖性下调TGFβ1、CTGF mRNA的转录和蛋白表达(P〈0.05或P〈0.01),减少细胞外基质的沉积(P〈0.05或P〈0.01)。结论糖肾清1号可通过抑制TGFβ1、CTGFmRNA转录和蛋白表达,抑制肾组织纤维化的发生发展。
Objective To observe the effect of Tangshenqing No.1 on the expression of cytokines mediating fibrosis in the renal issue. Methods Rats were divided into control group, model group, active-drug controlled group, and four treatment groups by Tangshengqing No.1, each designed as low-dose, mid-dose and high-dose groups. mRNA transcription level of TGFβ1 and CTGF were tested by RT-PCR and Western blot, histomorphological test was also conducted. Results Tangshenqing No.1 significantly down-regulated the expression of mRNA and protein (P0.05 或 P0.01), ECM sediment was also decreased (P 0.01 or P0.05). Conclusion Tangshenqing No.1 can curb the development of renal tissue fibrosis by regulating the mRNA transcription and protein expression of TGFβ1 and CTGF.
