以文蛤肉为原料,采用复合胰蛋白酶酶解,通过喷雾干燥工艺得到文蛤多肽粉.根据检验标准GB1519.3—2003食品急性毒性试验中最大耐受剂量法,对清洁级KM小鼠无毒害作用,作用后小鼠均健康无异常反应,解剖无明显病变,其皿0〉15g/kg体质量.对文蛤多肽粉组成成分进行分析,测定了其总氮、蛋白质、氨基酸、总糖、还原糖、脂肪、灰分及Na、K、Ca、Mg、Fe、Cu、Zn、Cd、Ag、Pb等金属元素的含量.应用基质辅助激光解吸附飞行时间质谱法(MALDI-TOF)分析了文蛤多肽粉分子量的分布范围.结果表明,该文蛤多肽粉中蛋白质、氨基酸和对人体有益的金属元素含量丰富,对人体有害的重金属含量都大大低于国家食品检验标准的安全剂量,氨基酸种类齐全;多肽分子量均小于5200u,其中分子量小于1000u的多肽占总多肽含量的69.03%.
Peptide product of Meretrix meretrix was prepared using the enzymatic catalysis of composite trypsin. On the national test criterion of GB1519.3 -2003 for the most enduring dosage in food urgent toxicity experiment, health product was not harmful to clear Kunming mice. After treated by the product,mice were normal in health and there was no illness in mice. The LD50 was higher than 15 g/kg weight. By analyzing the main component, metal content, amino acid composition and the range of polypeptide molecular weight, we found that in the Meretrix meretrix peptide, amino acid,saccharoid and metal elements are very rich. Meanwhile, heavy metals are very poor and much lower than the safe concentrations in national food test criterial. The categories of amino acids are complete. The polypeptide molecular weights are less than 5 200 u,polypeptide less in molecular weight is richer and polypeptide which molecular weight is less than 1 000 u is 69.03% of total polypeptide.
