目的:探讨比卡鲁胺对膀胱尿路上皮癌EJ细胞增殖、迁移和浸润功能的影响及其可能机制。方法:0,25,100μmol/L比卡鲁胺处理EJ细胞,MTT法检测细胞生长曲线;流式细胞PI单染法检测细胞周期;transwell细胞迁移实验法检测细胞迁移能力;transwell细胞侵袭实验法检测细胞侵袭能力;Western blotting检测雄激素受体(AR)、细胞周期蛋白D1、基质金属蛋白酶-2(MMP2)及瞬时受体蛋白家族成员-8蛋白(TRPM8)水平变化。结果:100μmol/L比卡鲁胺处理48 h后,EJ细胞增殖能力明显下降,细胞周期表现为G0/G1阻滞(P〈0.05);不同浓度(25,100μmol/L)比卡鲁胺处理48 h后,EJ细胞迁移及浸润能力明显降低,AR、Cyclin D1、MMP2和TRPM8蛋白表达明显降低。结论:比卡鲁胺能抑制膀胱癌EJ细胞增殖、迁移和浸润功能,其机制可能与抑制AR信号通路、降低相关蛋白表达有关。
Objective: To study the effect of bicalutamide on the proliferation,migration and invasion in bladder urothelial carcinoma EJ cells and to explore its mechanism.Methods: MTT method was used to examine the inbibitory of Bicalutamide on the proliferation of EJ cells.The cell cycle distribution influence of Bicalutamide on EJ cells was detected by flow cytometry.The inhibitory of Bicalutamide on the migtation and invasion of EJ was examined by Transwell chamber method.Western blotting was used to determine the expression of androgen receptor(AR),CyclinD1,MMP2,and TRPM8.Results: Bicalutamide inhibited the proliferation of EJ cells and induced the cycle arrest at G 0/G 1 phase.Bicalutamide suppressed the migration and invasion of EJ cells in a dose-dependent manner,and both the migration assays and invasion assays showed that the cell number were decreased at 25 and 100 μmol/L.Western blotting showed that the expression of AR,Cyclin D1,MMP2 and TRPM8 were significantly decreased after treatment with Bicalutamide.Conclusion: Bicalutamide can inhibit the proliferation,migration,and invasion of bladder urothelial carcinoma cell in vitro,and the mechanism refers to the androgen/AR pathway.
