为建立猪流感病毒(SIV)、猪圆环病毒2型(PCV-2)、猪繁殖与呼吸综合征病毒(PRRSV)单一或混合感染的快速检测方法,针对病毒保守区设计3对特异性引物用于扩增SIV、PCV-2与PRRSV基因的相应片段,并通过优化反应条件,建立了SIV、PCV-2、PRRSV三重PCR检测方法,其最低检测限度为10-4-10-5数量级。用三重PCR和单项PCR/RT-PCR检测40份临床病料,符合率为100%。提示建立的三重PCR检测方法能够对SIV、PCV-2、PRRSV单一或混合感染的临床样品进行快速鉴别检测,具有一定的临床应用价值。
According to the sequences reported in GenBank of virus conserved sequences,three pairs of primer were synthesized to amplify target sequences of SIV、PCV-2、PRRSV.Through the optimization of reaction conditions and the detection of clinical cases,the SIV,PCV-2and PRRSV triplex PCR was established.The results show that the triplex PCR had the minimum detection limit10-4-10-5.40clinical samples were detected by triplex PCR and single PCR/RT-PCR,the consistent rate was 100%.The results suggest that:The triplex PCR can be used for the detection of clinical samples with single or coinfections with three kinds of viruses.
