罗格列酮对PDGF-BB刺激的人气道平滑肌细胞增殖的影响

目的探讨罗格列酮对血小板源性生长因子BB(PDGF-BB)刺激的人气道平滑肌细胞(HASMCs)增殖的影响。方法组织块贴壁法培养HASMCs鉴定后,接种于96孔板,除空白对照组外,A、B、C、D组各孔在加入PDGF-BB前30min分别加入0、1、5、10μmol/L罗格列酮。MTS/PMS微量比色法检测各组HASMCs细胞A490吸光度,流式细胞仪检测细胞周期分布。结果A组A490值高于空白对照组,B、C、D各组均高于A组(P均〈0.05);与空白对照比较,单加入PDGF-BB后G0/G1期细胞比率降低,S期升高;用1、5、10μmol/L罗格列酮预处理的ASMCs在G0/G1期细胞比率逐渐升高,S期逐渐降低(P均〈0.05)。结论罗格列酮呈剂量依赖性抑制PDGF-BB刺激的HASMCs增殖。

英文摘要:

Objective To explore the effect of rosiglitazone on human airway smooth muscle cells (HASMCs) proliferation induced by platelet derived growth factor ( PDGF)-BB. Methods HASMCs were cultured by tissue adherence method. HASMCs were vaccinated in plate with 96 holes after identifition. 0,1,5,10 μmol/L rosiglitazone were separately added in A,B ,C,D groups except blank group for 30 minutes before adding PDGFBB. MTS/PMS microcolorimetry was applied to assess A490 absorption factor of HASMCs cell. In all groups, cell cycle distribution was detected by flowcytometry. Results A490 value in group A was higher than that of blank control, and lower in group B, C,D than that of group A (P all 〈 0.05 ). Cell ratio was lower in G0/G1 stage , and higher in S stage than blank control after adding PDGF-BB. The rati-os of ASMCs which were primed by 1,5,10 μmol/L rosiglitazone were upgraded in GO/G1 stage,and were depressed in S stage gradually(P all 〈 0.05). Conclusions Rosiglitazone has inhibitory influence on HASMC sproliferation induced by PDGF-BB in dose-dependment way.
