目的:探讨丹参素对高糖刺激培养的腹膜间皮细胞(HPMCs)分泌纤维连接蛋白(FN)和I型胶原(Col-I)的影响。方法:以高糖刺激培养HPMCs,分别加用浓度为10,5,2.5,1.25,0.625mg/L的丹参素进行干预,并选取浓度为10mg/L的丹参素干预0,12,24,48,72h;应用RT-PCR,ELISA法检测FN和Col-I蛋白及mRNA表达;RT-PCR、免疫荧光法检测血红素氧化酶-1(HO-1),内皮素-1(ET-1)蛋白及mRNA表达。结果:丹参素可显著降低高糖诱导的HPMCs细胞外基质FN和Col-I蛋白及mRNA表达,呈浓度依赖性及时间依赖性;丹参素可抑制高糖诱导的ET-1蛋白及mRNA表达,增加诱导型抗氧化酶HO-1蛋白及mRNA表达,呈浓度依赖性;丹参素不同时间组ET-1和HO-1蛋白及mRNA表达差异无统计学意义(P〉0.05)。结论:丹参素可减少高糖诱导的腹膜间皮细胞FN和Col-I的分泌,其作用机制可能与拮抗氧化应激状态有关。
Objective To investigate the possible role of Danshensu on fibronectin(FN) and collagen-I(Col-I) secretion induced by high glucose in human peritoneal mesothelial cells(HPMCs).Methods HPMCs were treated with high glucose and Danshensu at different concentrations.The mRNA expression of FN,Col-I,endothelin-1(ET-1),and heme oxygenase-1(HO-1) were examined by reverse transcription-polymerase chain reaction(RT-PCR).The protein expression of FN,Col-I HO-1 and ET-1 were analyzed by enzyme-linked immunosorbent assay or immunofluorescence method.Results The expression of protein and mRNA of FN and Col-I were attenuated by Danshensu in both dose-dependent and time-dependent manner.The mRNA and protein levels of ET-1 were decreased,and the mRNA and protein levels of HO-1 increased in the Danshensu groups in a dose-dependent manner compared with the high glucose group.The expression of ET-1 and HO-1 showed little difference in a time gradient of danshensu(P0.05).Conclusion Danshensu can protect HPMCs through inhibiting the expression of FN and Col-I induced by high glucose,which is related to the suppression of oxidative stress.
