目的:研究当飞利肝宁胶囊对大鼠非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)肝纤维化的预防作用。方法:雄性Wistar大鼠30只,随机分为正常组、模型组和治疗组。正常组大鼠喂食普通饲料,模型与治疗组大鼠给予高脂饲料,治疗组同时予当飞利肝宁胶囊灌胃。8周后,对模型与治疗组大鼠腹腔注射小剂量四氯化碳(carbon tetrachloride,CCl4),48小时后处死各组动物并取肝组织固定或冻存。观察各组大鼠肝组织天狼猩红染色结果,应用荧光定量RT-PCR和Western blot检测肝脏纤溶酶原激活物抑制因子-1(plasminogen activator inhibitor-1,PAI-1)、转化生长因子-β1(transforming growth factor beta 1,TGF-β1)、α-平滑肌肌动蛋白(alpha smooth muscle ac-tin,α-SMA)及I型胶原α1(collagen type I alpha 1,COL1A1)表达水平。结果:模型组大鼠肝组织出现广泛肝细胞脂肪变性,在肝小叶3区可见轻度窦周纤维化,而治疗组大鼠肝细胞周围无明显胶原沉积。模型组大鼠肝组织PAI-1、α-SMA、TGF-β1、COL1A1表达水平较正常组升高(P〈0.05),在治疗组大鼠肝脏中上述促纤维化基因的表达被显著下调(P〈0.05)。结论:当飞利肝宁胶囊对高脂饮食联合CCl4诱导的大鼠NAFLD肝纤维化具有预防作用,其机制可能与调控NAFLD进展中TGF-β1和PAI-1的表达从而抑制肝星状细胞活化、胶原增生及促进细胞外基质降解相关。
Objective: To study the effect of Dangfei Liganning capsules on liver fibrosis in nonalcoholic fatty liver disease(NAFLD) rats.Methods: Thirty male Wistar rats were randomly divided into normal group,model group and treatment group.The rats of normal group were fed with a standard diet,while those of model and treatment groups with high-fat diet,and the rats for treatment were gastrically perfused with Dangfei Liganning capsules.After 8 weeks,the rats of model and treatment groups were intraperitoneally injected with low dose of CCl4.Forty-eight hours later,the experimental rats were sacrificed and their livers were frozen or fixed for future use.The liver tissue slides were stained with Sirius red and observed through microscope.Hepatic levels of plasminogen activator inhibitor-1(PAI-1),transforming growth factor beta 1(TGF-β1),alpha smooth muscle actin(α-SMA),collagen type I,alpha1(COL1A1) and β-actin were measured by quantitative RT-PCR and Western blot.Results: Compared with normal group,the liver tissues of model rat showed extensive lobular hepatic steatosis,companied with mild peri-sinusoidal fibrosis in zone 3 area.No obvious collagen deposition was observed in the liver of treating rats.Dramatically increased mRNA level of PAI-1,TGF-β1,α-SMA and COL1A1,as well as the protein expressing level of PAI-1,were determined in the liver tissues of model rats(P < 0.05),while in the treatment group,the expression of these pro-fibrogenesis genes were down-regulated obviously(P < 0.05).Conclusion: Dangfei Liganning capsules could prevent liver fibrosis of NAFLD rats.The underlying mechanism might partially lie in inhibiting the activation of hepatic stellate cells and collagen production,as well as promoting the degradation of extracellular matrix,through regulating the expression of TGF-β1 and PAI-1 in NAFLD pathophysiology.
