目的对抗炎组分进行提取并对梣酮进行质谱鉴定;对白鲜皮粗提物及梣酮单体进行抗炎效果检测。方法采用电喷雾质谱技术对所分离梣酮进行鉴定;利用小鼠耳廓肿胀及小鼠腹腔毛细血管渗透性实验确定待试物抗炎活性。结果提出白鲜皮中梣酮的裂解规律;白鲜皮醇提物及梣酮单体各剂量组可显著抑制由二甲苯引起的小鼠耳廓肿胀,差异显著(P〈0.05),但在抑制醋酸致小鼠腹腔毛细血管渗透性增高方面,梣酮中、低剂量组效果明显。结论建立白鲜皮中梣酮快速分析鉴定方法,白鲜皮醇提物及梣酮单体具有很好的抗炎作用,但不同抗炎模型对抗炎效果表现具有差异性。
[ Objective ] To detect the anti-inflammatory effects of ethanol extracts and fraxinellone extracted and isolated from Cortex Dietamni. [Methods] To identify fraxinellone extracted and isolated from Cortex Dictamni by electrospray ionization mass spectrometry (ESI-MS), anti-inflammatory effects of ethanol extracts and fraxinellone were investigated by xylene-induced aruieal swelling in mice and the abdominal capillary permeability in mice. [ Results ] In this paper EI Mass Spectra of fraxinellone were measured, the results showed that ethanol extracts and fraxinellone were significantly (P 〈0.05) suppress the mouse auricle which caused by the xylene swelling. The medi- um-dosage and low dose group were increased significantly in abdominal capillary permeability in mice compared with control group. [ Conclusions ] A method using eleetrospray ionization tandem mass spectrometry(ESI-MSn) was established for analysis and identification of fraxinellone from Cortex Dietamni. The results showed that ethanol extracts and fraxinellone showed differences anti-inflammatory effect in variety of animal models of anti-inflammatory.
