为了探究浙江曹娥江下游可培养嗜盐细菌的分布特征,通过平板分离筛选技术研究了该水系中可培养嗜盐细菌的种类及数量.研究结果表明,在37℃和50℃下,通过3种常用嗜盐菌分离培养基,共获得可培养嗜盐细菌54株.对其16SrRNA基因序列进行分析可知,这些细菌由γ-变形菌纲(γ-Proteobacteria)(19株)和芽孢菌纲(Bacilli)(35株)中的8类微生物种群组成,隶属于盐单胞菌属(Halomonas)、枝芽孢杆菌属(Virgibacillus)、芽孢杆菌属(Bacillus)和葡萄球菌属(Staphylococcaceae).对这8类种群的α-多样性指数进行分析,结果表明其多样性不高且均匀度低,优势种群明显,尤以表皮葡萄球菌(Staphylococcus simulans Z482)居多,占整个群落个体的38.89%.这与类似环境中嗜盐细菌的组成分布有较大的差异,可能与河流的位置以及当地生产生活废水的大量排入有关.
Stalinization phenomenon of Cao'e River is increasingly serious in Zhejiang province in recent years.In order to investigation distribution characteristics of halophilic bacteria,the species and numbers of halophilic bacteria cultured were screened and isolated by plating method.The result showed fifty four halophilic bacteria were isolated from the lower Cao-E River in Zhejiang province by the method of plate isolation.Based on 16 SrRNA gene sequence andα-diversity index analysis,the bacteria were divided 8kinds of microbial populations ofγ-Proteobacteria(contained 19strains)and Bacilli(contained35strains),which belong to four different genera,i.e.,Halomonas,Virgibacillus,Bacillus and Staphylococcaceae.The halophilic bacteria in this region had a lower population diversity and evenness,but its dominant population was obvious,especially the majority of Staphylococcus simulans Z482 accounted for38.89% of the entire community individuals.Compared with the similar habitats,the halophilic bacteria of the lower Cao-E River has different composition and distribution characteristics,maybe it was related to the mass discharge of local production and living waste water.
