目的观察瓜子金皂苷己(polygalasaponin F,PGSF)对氧糖剥夺/复供(oxygen-glucose deprivation and reperfusion,OGD/R)所诱导的PC12细胞凋亡的作用及其机制。方法以连二亚硫酸钠合并无糖Earle’s液造成氧糖剥夺,继而恢复为完全培养基以建立体外OGD/R模型,以Hoechst33342/PI双染及流式细胞术观察细胞受损和凋亡情况;以JC-1荧光染色法测定细胞线粒体膜电位(mitochondrial membranepotential,MMP);以Western blot法检测凋亡相关蛋白的表达。结果 PGSF可明显改善细胞形态并降低细胞受损和凋亡百分率,抑制MMP水平的降低,增加Bcl-2/Bax表达比值。结论 PGSF对OGD/R诱导的PC12细胞凋亡具有明显抑制作用,机制与其调节Bcl-2和Bax的表达及稳定MMP有关。
Aim To study the effect of Polygalasapo- nin F (PGSF) on apoptosis of PC12 cells induced by oxygen-glucose deprivation and reperfusion (OGD/R) and its mechanism. Methods The OGD/R model was established by first exposing PC12 cells to sodium dithionite (Na2S2O4 ) in glucose-free Earle' s solution and then by changing the culture into complete mediums, with the cell injury and apoptosis analyzed by Hoechst 33342 / PI dual staining and flow eytometry assay. The mitochondrial membrane potential (MMP) was determined by JC-1 staining, and the expression of Bcl-2 and Bax was detected by Western blot. Results PGSF could significantly improve the morphology of injured PC12 cells and reduce the percentage of apoptotic cells, reverse the decrease in MMP and up-regulate the ratio of Bcl-2/Bax proteins. Conclusion PGSF can markedly inhibit the OGD/R-induced apoptosis of PC12 cells by regulating the expression of Bcl-2 and Bax proteins and maintaining the normal MMP.
