丝氨酸/苏氨酸激酶33(serine/threonine kinase 33,STK33)是近年来倍受生物学界关注的一种蛋白因子。它并非普遍表达,且在大多数组织中表达水平较低。虽然STK33属于钙离子/钙调素依赖性蛋白激酶(Ca^2+calmodulin-ependent protein kinase,CAMK)家族,但是STK33在表达模式上与某些CAMK家族亚型成员有一定的差异。STK33可能通过特异性磷酸化波形蛋白,参与中间丝细胞骨架解聚的动态变化,影响细胞的结构和一系列重要的功能,进而参与肿瘤的发生和发展过程。STK33在细胞信号转导过程中,尤其是与癌基因存在交互依赖作用时,对调控肿瘤增殖起特殊作用。STK33参与依赖于Ras癌基因的多种肿瘤细胞的“合成致死”作用。STK33有可能作为新靶点,在肿瘤的分子靶向治疗中发挥重要作用。
In recent years, STK33 (serine/threonine kinase 33), a novel protein factor, is mostly concerned in biological field. It displays a non-ubiquitous expression and the expression level is lower in most tissues. STK33 belongs to the CAMK (Ca^+2+/calmodulin-dependent protein kinase) family, while there are certain differences in the expression patterns between STK33 and other members of CAMK family. STK33 may participate in the dynamic changes in depolymerization of intermediate filament cytoskeleton as well as affect the cell structure and a series of important functions by specific phosphorylation of vimentin, thereby involving in the initiation and development of tumor. In the process of cellular signal transduction, especially in the existence of interplay dependent on oncogenes, STK33 plays a special role in the regulation of tumor cell proliferation. STK33 participates in the "synthetic lethal" process of tumor cells which depend on the Ras oncogene. STK33 may possess an important role in molecular-targeted therapy for tumors, as an attractively novel target.
