目的研究中药不同分期治疗对SD大鼠骨折愈合过程中BMPs表达的影响,探讨中药治疗骨折的合理分期方法。方法于SD大鼠胫骨上1/3段前部中点造成2mm×1.5mm的骨质缺损。246只雄性SD大鼠随机取6只作为正常对照组,其余于造模后随机平均分为4组,分别给予相应干预措施:三期组,三期治疗;二期组,二期治疗;一期组,一期治疗;模型组,生理盐水。每组于术后不同时间点分别各处死6只,常规标本处理、组织学检查和免疫组化检测标本中的BMPs,进行图像分析。结果(1)各时间点BMPs表达平均阳性细胞数(PCN)及平均光密度(MOD)值的总体平均值和多个时间点的PCN与MOD:二期组〉三期组〉一期组〉模型组,P值〈0.05。(2)BMPs表达出现芝个高峰,二期组高峰期持续时间显著长于其它组。(3)二期与三期组BMPs表达峰值水平显著高于一期组和模型组,P值〈0.05,二期组有高于三期组的趋势。结论中药能显著促进SD大鼠骨折愈合过程中BMPs的表达,中药治疗骨折进行分期辨证治疗是必要的,二期治疗可能是其最佳分期方法。
Objective To probe the reasonable phasing approach in treatment of fracture with TCM. Methods A 2 mm- wide - and - 1.5 mm- deep defect was bilaterally made as fracture healing model in the middle of the anterior part of the up - shaft of tibia of a rat. First ,6 male SD rats were randomly taken into the normal control group, postoperatively, the other 240 were randomly divided into four groups on average and given corresponding intervention : three - phase treatment group, two - phase treatment group, one - phase treatment group and model control group. On different days after the operation, rats were killed respectively. Specimens were prepared and examined histologically in routine. The expression of BMPs was detected by immunohistochemieal staining and quantitatively analyzed. Results ( 1 ) Group mean MOD (mean optical density) and positive cell number of BMPs expression in every group at deferent time points and those of more time points : two - phase treatment group 〉 three - phase treatment group 〉 one - phase treatment group 〉 model control group, P 〈 0. 05. ( 2 ) BMPs expression presented 2 peaks, and the lasting time of peak - stage in 2 - phase treatment group was considerably longer than that in other groups. ( 3 ) The top level of peak of BMPs expression in both two -phase and three -phase treatment group were significantly higher than that in the other two ,P 〈 0.05. Conclusions TCM is able to greatly promote BMPs expression in fracture healing of SD rats, and it's very necessary to treat fracture with TCM by phases, and two - phase treatment was the most reasonable phasing way.
