目的观察β-榄香烯对胃癌MGC803细胞凋亡的影响,并探讨其机制c用不同浓度β-榄香烯作用于MGC803 24~48 h,MTT法检测细胞活力,流式细胞术测算细胞凋亡率,Western blot法检测MGC803中的磷酸化细胞外信号调节激酶(p-ERK)及总ERK。结果随着药物浓度增加,细胞存活率逐渐降低(P均〈0.05),细胞凋亡率明显增高(P均〈0.05);随着药物浓度增加、作用时间延长,MGC803中p-ERK表达量显著下降(P均〈0.05)。结论β-榄香烯可诱导胃癌MGC803凋亡,其机制可能与抑制细胞ERK通路活性有关。
Objective To investigate the effect and mechanism of β-elemene on apoptosis in human gastric cancer MGC803 cells.Methods Human gastric cancer MGC803 cells were treated with different concentrations of β-elemene for 24~48 h,and the cell viability was measured by MTT assay,apoptosis was evaluated by flowcytometry following propidium iodide(PI) staining.Western blot was employed to detect the expression of phosphor-extracellular signal-regulated kinases(ERK) and total ERK after the cells were treated with β-elemene.Results β-elemene inhibited the viability of MGC803 cells in a dose-dependent manner.Further study showed that β-elemene induced apoptosis and inhibited ERK activity.Conclusion β-elemene can induce apoptosis in human gastric cancer MGC803 cells probably by inhibiting ERK signaling pathway.
