目的考察中药当归和川芎煎液的化学成分差异,以探讨其药效差异的物质基础。方法采用UPLC-QTOFMS技术对市场上正规销售的当归和川芎饮片进行比较。数据统计分析采用主成分分析和正交偏最小二乘法判别分析。成分鉴别采用与标准品比较、基于自制自动化预鉴别软件的一级质谱精确质荷比分析和二级质谱碎片分析。结果当归和川芎煎液化学成分差异明显;三糖以及可能含有丁烯基苯酞结构的化合物等在当归、川芎煎液中含量差异明显;阿魏酸含量差异不突出。结论川芎煎液三糖含量高于当归煎液,另外发现多种有明显差异的LC-MS信号,为探讨当归、川芎药效差异提供了参考。

英文摘要:

Objective To investigate the chemical difference between decoctions of ANGELICAE SINENSIS RADIX and CHUANXIONG RHIZOMA in order to research the material basis of pharmacodynamic differences.Methods Samples from legal markets were compared using ultra-performance liquid chromatography-quadrupole timeof-flight mass spectrometry( UPLC-QTOF-MS). Data were statistically analyzed by principal component analysis and orthogonal partial least-squares to latent structures discriminate analysis. Constituents identification involved comparision with reference compounds,accurate m / z value analysis of TOF-MS data based on in-house software for automatic tentative identification,and QTOF-MS / MS fragment analysis. Results The contents of such compounds as trisaccharide and those that may contain butylidenephthalide structure were significantly different between the two types of decoctions. Difference in contents of ferulic acid was not prominent. Conclusion The content of trisaccharide in CHUANXIONG RHIZOMA may be higher than in ANGELICAE SINENSIS RADIX. Various LC-MS signals which are significantly different between ANGELICAE SINENSIS RADIX and CHUANXIONG RHIZOMA are identified. This research provides reference for the study of chemical composition and efficacy between the two kinds of decoctions.
