目的研究人脐带血间充质干细胞(mesenchymalstemcells,MSC)移植对兔局灶性脑缺血再灌注损伤神经生长因子(NGF)和脑源性神经营养因子(BDNF)表达及皮质神经元凋亡的影响。方法健康雄性新西兰大白兔32只,随机分为假手术组、缺血再灌注组、MSC组、生理盐水组,每组8只。于3、7、14d对兔神经功能进行评分,并测定血清NGF、BDNF含量变化,14dTUNEI。法检测缺血灶周围大脑皮质细胞凋亡情况。结果Msc组缺血再灌注7、14d神经功能缺损评分明显低于缺血再灌注组和生理盐水纽(P〈0.05)。缺血再灌注纽和生理盐水组3、7、14d血清NGF和BDNF含量较假手术组明显升高(P〈O.05)。MSC组3、7、14d血清NGF和BDNF含量明显高于缺血再灌注组和生理盐水组,差异有统计学意义(P〈o.05)。与假手术组比较,缺血再灌注组神经细胞凋亡指数明显升高(P〈0.05);MSC组神经细胞凋亡指数较缺血再灌注组和生理盐水组明显降低,差异有统计学意义(Pd0.05)。结论MSC移植可促进脑缺血再灌注兔的神经功能缺损恢复,增加血清NGF、BDNF分泌,并可抑制脑缺血再灌注诱导的神经细胞凋亡。
Objective To study the effect of mesenchymal stem cells (MSC) from human umbilical cord blood on NGF and BDNF expression and apoptosis of cortical neurons in rabbits following focal cerebral ischemia/reperfusion (I/R) injury. Methods Thirty-two healthy New Zealand white rabbits were randomly divided into sham group, I/R group, MSC group and saline group (8 in each group). Their nerve function was scored and their serum NGF and BDNF levels were measured on days 3,7 and 14, respectively, after reperfusion. Apoptosis of cerebral cortical cells was assyed by TUNEL on day 14 after reperfusion. Results The nerve function defect score was significantly lower in MSC group than in I/R group and saline group on days 7 and 14 after reper- fusion (P(0.05). The serum NGF and BDNF levels were significantly higher in I/R group,saline group and MSC group than in sham group on days 3,7 and 14 after reperfusion (P〈0. 05). The apoptosis index of MSC was significantly higher in I/R group than in sham group and significantly lower in MSC group than in I/R group and saline group (P〈〈0.05). Conclusion MSC transplan- tation can promote the recovery of nerve function defect, increase the secretion of NGF and BDNF,and inhibit the apoptosis of nerve cells in rabbits following focal cerebral I/R injury.
