目的研究miR.152分子对JEG-3细胞中HIA—G表达水平的调控。方法在滋养细胞肿瘤细胞系JEG-3细胞中分别转染pre-miR-152(实验组),Cy3标记的pre—miRNA—control(阴性对照),空白对照(NC)为未转染的JEG-3细胞。转染48h、72h分别进行RT.PCR与Westernblot实验检测细胞中miR-152、HLA—GmRNA与蛋白的表达水平。结果RT-PCR结果显示在细胞中转染pre—miR-152分子后miR-152的表达显著增高(P〈0.05);转染后HLA—GmRNA的表达水平在各组之间差异无统计学意义(P〉0.05)。Westernblot结果显示与对照组相比,实验组的HLA—G蛋白表达水平显著降低(P〈0.05)。结论miR-152可能在翻译水平抑制HLA-G的表达。
Objective To investigate the effect of miR-152 on the expression of HLA-G in JEG-3 cells. Methods The trophoblastic tumor JEG-3 cells were transfeeted with pre-miR-152(as test group),pre-miRNA-control labeled with dye Cy3 (as negative control) , and with nothing( normal control, NC) , respectively. 48 and 72 hours after transfction, ceils were used for RT-PCR and Western blot analysis to investigate the expression of miR-152 mRNA and HLA-G mRNA, HLA-G protein, respectively. Resuits After pre-miR-152 transfection, RT-PCR analysis showed that the transfection was effective( P 〈 0.05 ) , whereas no significant changes were observed in the mRNA level of HLA-G. However, the cells transfected with pre-miR-152 significantly decrease the HLA-G protein level(P 〈 0.05 ). Conclusion The miR-152 can down-regulate the expression of HLA-G by inhibiting the translation process.
