目的:观察C57BL/6小鼠肝脏中NK细胞、T细胞和NKT细胞的含量、亚型和细胞因子产生能力。方法:分离正常C57BL/6小鼠肝脏的淋巴细胞,通过细胞表面分子染色以及流式细胞术(FCM)检测NK1.1^+NK细胞、CD3^+T细胞和CD3^+NK1.1^+NKT细胞的含量及其表面CD4、CD8分子的表达情况;细胞经过PMA+Ionomycin刺激后,应用细胞内细胞因子染色并结合FCM检测NK1.1^+NK细胞、CD3^+T细胞和CD3^+NK1.1^+NKT细胞产生IFN-γ、IL-4、IL-9和IL-17细胞因子的情况。结果:肝脏中NKT细胞的含量为(21.51±13.02)%,高于NK细胞和T细胞(P〈0.05)。NK细胞以CD4^-CD8^-亚群为主,T细胞和NKT细胞均以CD4^+细胞亚群为主。NKT细胞分泌的IL4的比例高于NK细胞和T细胞(P〈0.05)。T细胞分泌IL-17的能力明显强于NK细胞和NKT细胞(P〈0.05),IL-9^+细胞在三种细胞中含量不高(〈0.03%)。结论:C57BL/6小鼠肝脏中NK细胞、T细胞和NKT细胞含量、表型和功能方面可能存在明显的差异。
Objective:To observe the content, subtype and production ability of NK, T and NKT cells in the livers of C57BL/6 mice. Methods: The lymphocytes were isolated from the livers of normal C57BL/6 mice. The surface molecule staining and flow cytometry ( FCM ) were used to detect the contents of NK1.1^+ NK, CD3^+ T and CD3^+ NKI. 1^+ NKT and expressions of CD4 and CD8 on their surfaces. After stimulation of PMA^+ Ionomycin, the intracellular cytokine staining combined with FCM was used to detect the productions of IFN-γ, IL-4, IL-9 and IL-17 by NK1. 1^+ NK, CD3^+ T and CD3 + NK1. 1 + NKT cells. Results: In the liver, the content of NKT cells was ( 21.51 + 13.02 ) %, which was significantly higher than that of the NK and T cells ( P 〈 0.05 ). The CD4 - CD8 - subgroup was mainly in the NK cells, while the CIM^+ subgroup was mainly in both the T and NKT cells. The proportion of IL-4 secreted by NKT cells was higher than that of NK and T cells ( P 〈 0. 05 ). The IL-7 secretion ability of T cells was significantly higher than that of NK and NKT cells ( P 〈 0.05 ). However, the content of IL-9^+ cells was not high in these three cells ( 〈0.03% ). Conclusion: There might be significant difference in content, phenotype and function in NK cells, T cells and NKT cells from the livers of C57BL/6 mice.
