目的观察二仙汤对卵巢切除大鼠胰岛素敏感性的影响及其机制。方法取SD大鼠40只,随机分为5组,每组8只,即假手术组,模型组,二仙汤低、高剂量组(30,60mg·kg-1),雌激素组(0.01mg·k-1)。灌胃给药连续治疗14周后,检测各组大鼠血糖、血清胰岛素、糖耐量,并计算胰岛素抵抗指数;应用CT检测各组大鼠腹腔内脂肪及皮下脂肪的相对量;应用qPCR技术检测各组大鼠腹腔内脂肪中MCP-1、脂联素的mRNA相对量。结果各组大鼠空腹血糖水平差异无统计学意义;模型组大鼠血清胰岛素水平显著高于假手术组;模型组大鼠胰岛素抵抗指数显著高于假手术组,二仙汤治疗后可降低卵巢切除大鼠胰岛素抵抗指数;cT结果显示,卵巢切除大鼠腹腔内脂肪含量显著高于假手术组,二仙汤治疗后脂肪含量显著下降;二仙汤治疗后能改善卵巢切除大鼠糖耐量并降低胰岛素抵抗指数;卵巢切除后大鼠腹腔内脂肪中MCP一1mRNA水平表达升高而脂联素的mRNA表达水平下降,二仙汤治疗后脂肪MCP一1的mRNA表达水平下降而脂联素的mRNA表达水平升高。结论二仙汤可提高卵巢切除大鼠胰岛素敏感性,这可能与其改善卵巢切除大鼠腹腔内脂肪内分泌功能有关。
Objective To observe the effects of Erxian Decoction (EXD) on insulin sensitivity in ovariectomized rats and to explore the mechanism. Methods Forty SD rats were randomly divided into five groups, namely sham operation group, ovariectomy group, low- and high-dose EXD groups(30, 60 mg·kg-1) , estrogen treatment group(0.01 mg·kg-l). After oral use of the corresponding medicine for 14 weeks, serum glucose, serum insulin, and glucose tolerance were determined, and the insulin resistance index was calculated. The relative levels of intra-abdominal fat and subcutaneous fat were tested by CT. The levels of monocyte chemoattractant protein-1 (MCP-1) and adiponectin in intra-abdominal fat were determined by qPCR. Results The glucose levels were not significantly different among the groups. The insulin level in ovariectomy group was significantly higher than that in the sham operation group. The treatment of EXD had an effect on decreasing the insulin resistance index of the ovariectomized rats. The results of CT examination showed that the ovariectomized rats had higher intra-abdominal fat level than the sham operation group, and EXD treatment could decrease the fat level, and improve the glucose tolerance. The mRNA level of MCP-1 was significantly higher while the mRNA level of adiponectin was significantly lower in intra-abdominal fat of ovariectomized rats as compared with the sham operation group, and EXD treatment could decrease the MCP-1 mRNA expression level and increase the mRNA levels of adiponectin in intra-abdominal fat of ovariectomized rats. Conclusion EXD treatment exerts certain effect on improving insulin sensitivity in ovariectomized rats, and the possible mechanism is related with the improvement of the endocrine function of intra-abdominal fat.
