目的建立UPLC法同时测定桑白皮Mori Cortex30%、50%、80%乙醇组分中桑皮苷A、桑辛素M-6,3'-O-β-D-二葡萄糖苷、5,7-二羟基香豆素、东莨菪亭、桑辛素M、桑辛素M-3'-O-β-D-葡萄糖苷、桑黄酮G、桑辛素的含有量。方法分析采用Dionex AcclaimTMRSLC 120 C18色谱柱(2.1 mm×100 mm,2.2μm);以0.1%乙酸水溶液(A)-甲醇(B)(30%乙醇)、0.1%磷酸水溶液(A)-甲醇(B)(50%乙醇)、0.1%磷酸水溶液(A)-乙腈(B)(80%乙醇)为流动相,梯度洗脱;体积流量0.3 m L/min;柱温20℃;检测波长305 nm(30%乙醇)、315 nm(50%乙醇)、280 nm(80%乙醇)。结果 8种成分均呈良好的线性关系(r〉0.999 0),平均加样回收率95%~105%,RSD均小于5%。结论该方法准确简便,灵敏度高,适用于桑白皮中化学成分的测定。
AIM To establish a UPLC method for simultaneously determining the contents of mulberroside A, moracin M-6, 3'-di-O-13-D-glucopyranoside, 5, 7-dihydroxycoumarin, scopoletin, moracin M, moracin M-3'-O- [3-D-glucopyranoside, kuwanon G and moracin in 30% , 50% and 80% ethanol extracts of Mori Cortex. METH- ODS The analysis was performed on Dionex AcclaimTM RSLC 120 Cls column (2. 1 mmx 100 ram, 2.2 μm) , mobile phases were 0. 1% acetic acid aqueous solution (A) -methanol (B) (30% ethanol), 0. 1% phosphoric acid aqueous solution (A) -methanol (B) (50% ethanol) and 0. 1% phosphoric acid aqueous solution (A) - acetonitrile (B) (80% ethanol) for gradient elution, flow rate was 0.3 mL/min, column temperature was main- tained at 20 ~C, and detection wavelengths were set at 305 nm (30% ethanol), 315 nm (50% ethanol) and 280 nm (80% ethanol). RESULTS These eight constituents showed good linear relationships (r 〉 0. 999 0), whose average recoveries were 95% - 105% with the RSDs of less than 5%. CONCLUSION The method is ac- curate and simple with high sensitivity, which is suitable for the determination of chemical constituents in Mori Cortex.
