目的对Piezo2性质、结构和功能进行预测分析,并检测其在肠道的表达。方法利用Protparam程序预测人Piezo2蛋白基本理化性质;TMHMM Serve 2.0预测跨膜结构域;ProtComp Version 9.0分析亚细胞定位;用SOPMA程序预测其二级结构;SWISS-MODEL服务器生成其三维模型。免疫组织化学染色确定Piezo2在肠道组织的表达位置,以实时定量荧光PCR检测Piezo2在不同肠段的表达量及与Epac1表达的相关性。结果 Piezo2蛋白分子式为C14539H22436N3662O4087S130,相对分子质量为318.092×103,等电点pI=5.82。Piezo2蛋白存在35个跨膜结构域,并有多个蛋白修饰位点。免疫组织化学染色发现Piezo2蛋白表达于肠道上皮与神经节,其在结肠表达水平较回肠丰富(P〈0.01),其中结肠Piezo2与Epac1在mRNA水平表达呈正相关(r2=0.65,P〈0.01)。结论 Piezo2在结肠表达丰富,可能通过Epac1通路参与肠道机械力感知与调控。
Objective To predict and analysis the physical and chemical properties as well as the structure and function of Piezo2,and to detect the expression of Piezo2 in gut.Methods Physical and chemical properties of Piezo2 were predicted with Protparam.Transmembrane domain was predicted with TMHMM Serve 2.0.Subcellular location was analyzed with ProtComp Version9.0.Protein secondary structure was predicted with SOPMA program.Three-dimensional model was created with SWISS-MODEL.Location of Piezo2 in intestine was analyzed with immunohistochemical staining and RT-PCR was performed to compare the expression in different segment of intestine and correlation to Epac1.Results Molecular formula of Piezo2 was C14539H22436N3662O4087S130with molecular weight of 318.092×103 and isoelectric point of 5.82.Piezo2 had 35transmembrane domains and many post-translational modification sites.Piezo2 existed in intestinal epithilial tissue and ganglion cells.Expression of Piezo2 in colon was much more abundant than that in small intestine(P〈0.01)and positive correlated to Epac1(r2=0.65,P〈0.01).Conclusion Colon is rich in Piezo2.Piezo2 might play a key role in function regulation of intestinal epithelium and enteric nervous system by Epac1 pathway.
