以5个亚麻品种陇亚10号、张亚2号、CDC-scroll、低亚麻酸及不育系1S为试验材料,对不同基因型亚麻下胚轴的不定芽诱导进行研究。确定了5个亚麻品种最适的种子消毒处理为75%乙醇消毒0.5min、1%次氯酸钠消毒10min。筛选出亚麻下胚轴不定芽诱导分化的最适培养基为MS培养基附加0.5mg/L6.BA和0.02mg,/LNAA。讨论了不同基因型的亚麻下胚轴不定芽诱导分化及其不定芽生根之间的差异,最终得到陇亚10号的下胚轴不定芽诱导分化率及不定芽的生根率最高.宜用来做后续的研究工作。
Five flax varieties (Longya No. 10, Zhangya No. 2, CDC-seroll, Diya, 1S )were used as experimental materials to study the adventitious shoot induction in different genotypes of flax hypocptyl. The results showed that the optimal disinfection treatments for five flax seeds were :70% alcohol solution for 0.5min, and then 1.0% sodium hypochlorite solution for 10min. The optimal culture medium of adventitious shoot was MS + 0.5mg/L 6-BA + 0.02mg/L NAA. The differences between flax hypocotyl adventitious shoot induction and adventitious shoot rooting of different genotypes were discussed. It was concluded that Longya No. 10 had the highest adventitious shoot induction rate and adventitious shoot rooting rate, and it's easy to do the follow-up research work, which provide foundation for the tissue culture of flax genetic transformation.
