目的探讨正常3T3-L1脂肪细胞和胰岛素抵抗(IR)脂肪细胞微小RNA(miRNAs)的差异性表达,并进行初步分析。方法采用高糖(25 mmol/L葡萄糖)/高胰岛素液(1μmol/L)处理3T3-L1脂肪细胞24h,通过葡萄糖摄取实验判断IR脂肪细胞模型的建立;接着通过miRNA芯片技术,检测脂肪细胞和IR脂肪细胞中miRNAs的表达,并对其分析;随后借助生物信息学方法,对显著变化的2个miRNAs(miR-320和miR-21)寻找并筛检其可能调控的靶基因。结果共获得79个IR相关miRNAs(29个低表达,50个高表达,其中miR-320显著上调,miR-21显著下调),且筛选出与IR或糖尿病相关的靶基因共13个。结论获得了3T3-L1脂肪细胞和IR脂肪细胞miRNAs的差异性表达谱,为进一步研究这些miRNAs在IR和糖尿病的发病机制中的作用奠定了基础。
【Objective】 To investigate differential expression profiles of miRNAs between normal 3T3-L1 adipocytes and insulin-resistant adipocytes(IR-adipocytes),and analyze their functions preliminarily.【Methods】 IR-adipocyte model was induced with high glucose(25 mmol/L) and high insulin(1μmol/L) in combination(high glucose/ insulin) for 24 hours.To identify the success of IR-adipocyte model,-glucose uptake was detected.Then the profile alteration of miRNAs was examined and analyzed by miRNA microarray analysis.Subsequently,the potential target genes of the miR-320 and miR-21 were predicted and screened by means of Bioinformatics.【Results】 According to a microarray-based screening,79 insulin resistance related miRNAs were obtained,including 50 highly-expressed miRNAs and 29 lowly-expressed ones.Among these miRNAs,miR-320 was markedly up-regulated,while miR-21 down-regulated;and 13 potential insulin-resistant or diabetes-related target genes of the two miRNAs were predicted by informatics analysis.【Conclusions】 MiRNA expression profiles in 3T3-L1 adipocytes and IR-adipocytes have been obtained,so as to lay necessary basis for further research on the function of miRNAs in IR and diabetes.
