为了建立新型钙通道拮抗剂MN9201的高效液相色谱(HPLC)手性拆分方法,采用DaicelOJ-H手性固定相色谱分析柱(250mm×4.6mm,5μm),优化色谱条件,拆分MN9201消旋体。建立的HPLC手性拆分条件为:用DaicelOJ-H手性分析柱,以正己烷-异丙醇-三乙胺(91:9:0.1,V:V:V)为流动相,流速为0.5mL/min,检测波长为237nm,MN9201对映体可达到基线分离。本研究为MN9201单一对映异构体的分离、制备及含量测定等提供了简单、快捷的方法。
To develop an enantiomeric separation method for MN9201--a dihydropyridine calcium channel blocker by high performance liquid chromatography (HPLC) , the enantiomers of MN9201 were separated on Daicel OJ-H column, a chiral stationary phase column by optimizing conditions of chromatogram. The chromatographic separations were performed using a OJ-H, 5μm, 250 mm× 4.6 mm column and using a mobile phase, hexane : isopropanol : triethylamine (91 : 9 : 0.1 ,V : V : V). The flow-rate of the mobile phase was 0.5 mL/min. The samples were monitored with UV detection at 237 nm. Under the optimized conditions, the enantiomers of MN9201 were well separated from the baseline. The proposed method is easy and accurate. It is reliable in the separation, preparation and quantification of MN9201.
