目的:验证经亲和淘选噬菌体展示随机十五肽库获得的与ICAM-1高亲和力结合的目的肽的生物功能。方法:在动物体外用ELISA法检验目的肽与ICAM-1的结合。在动物体内,先荧光标记目的肽,后将目的肽注射人小鼠炎症模型体内,肾脏组织冰冻切片定位荧光物质。结果:ELISA法验证目的肽与人ICAM-1呈浓度依赖性结合。在动物体内,重新荧光标记的目的肽具有向高表达ICAM-1的炎症部位特异性聚集的功能。结论:此目的肽具有与组织中高表达的ICAM-1特异性结合的生物功能,说明此目的肽可尝试作为以ICAM-1为靶的“肽导向药物”的前导肽。
Objection:Verify the biological functions of ICAM-1-binding peptide which had been screen from phage-displayed random 15-peptide library. Methods:in v/tro,to prove the target peptide conbines with ICAM-1 by ELISA analysis. In vivo, the target peptide was labeled with fluorescence and was injected in mouse inflammatory model bodys to orientate fluorescence by freezing tissues slices. Result:The target peptide combines with ICAM-1 related with concentration. The target peptide could accumulate toward ICAM-1 in vivo. Conclusion:The target peptide can conbine with ICAM-1 specially. It suggested that the target peptide might be used as the guiding peptide of "peptide guided drug" to inflammatory site with ICAM-1 expression in the future.
