冬虫夏草是一种名贵的强壮滋补中药,但由于其严格的寄生性和特殊的生长地理环境,天然虫草资源越来越少,冬虫夏草的开发利用只有走人工培养之路.经研究蛹虫草的培养时间与菌丝体生产量的关系,虫草多糖的提取以及虫草多糖的测定,结果显示培养6d的菌丝产量最高,发酵时间对菌丝生长量的研究有重要意义,它可以控制大规模发酵生产大周期.多糖含量的测定用硫酸-苯酚法.经实验显示2次浸提比1次浸提的得率要高,摇瓶菌丝体的粗多糖得率为25.0%,精多糖得率为10.15%,均比已报道的虫草多糖得率高,这为人工控制大规模发酵生产大周期提供科学依据.
Cordyceps sinensis is a rare Chinese traditional medicinal herb with strong tonic function.Because of its strict parasitic and special growth environment,natural cordyceps sinensis is getting less and less.Artificial culture is the only way to exploitation and utilization of Chinese caterpillar fungus.By studying the relationships between culture time and mycelium production of cordyceps sinensis,the extraction and the mensuration of cordyceps polysaccharide,the results show that six days culture can get the highest amount of hypha.It is important to investigate the fermentation time on hypha growth because it can control the fermentation cycle in large-scale.The polysaccharide content is determined with sulfuric acid-phenol method.The results show that extraction the polysaccharide twice is better than only once,25.0%of the coarse polysaccharides or 10.15%of the pure polysaccharides is able to yield from shake flask mycelial,which are both higher than those reported from other researchers.These data can provide scientific basis for the manual control on large-scale fermentation production.
