红树林内生真菌732#可产生具有抗癌活性的新化合物enniatinG。出于对enniatin G的兴趣,通过ITS法测序分析,对732#种属进行了鉴定,得出732#属于Fusarium属真菌。为了增加732#的代谢产物量,以菌丝质量浓度(干质量)和enniatin质量浓度为检测指标,用正交法确定了适合732#生长的最佳碳源、氮源及温度,再用单因素变换法对732#培养条件进行了优化,得出,在含W=2.5%葡萄糖,W=0.8%蛋白胨,起始值为pH6,20%人工海水的GTY培养基中发酵10d,菌丝质量浓度(干质量)和enniatin质量浓度可达最大值。
The endophytic fungus 732# , isolated from the mangrove in the South China Sea, with anticancer novel Enniatin G, was characterized by ITS sequence analysis and phylogenetic analysis, which showed that the 732# belongs to the genus of Fusarium sp. To increase the content of the metabolites of the 732#, orthogonal test was used to select the optimum carbon sources and nitrogen sources with dry mycelium weight as observed objects; The incubation conditions for the growth and enniatin production of the Fusarium 732# were studied through changing the concentrations of the carbon sources and nitrogen sources, salt concentrations, and the initial pH values with enniatins and dry mycelium weight as observed objects. Under the optimal fermentation conditions, in GTY medium containing 25. % glucose, 0. 8% tryptone , pH 6, 20 % artificial seawater and 10 days incubation , the maximal dry weight and enniatin yield were obtained.
