目的:探讨右旋一叶萩碱对小鼠黑色素瘤B16细胞增殖和细胞周期的影响及其作用机制。方法:右旋一叶萩碱作用于小鼠黑色素瘤细胞B16,MTT法检测细胞增殖;流式细胞术检测细胞周期。结果:右旋一叶萩碱以时间和浓度依赖的方式抑制B16细胞生长;药物作用72 h IC50为63.34μmol/L;右旋一叶萩碱诱导B16细胞凋亡并显著减少S期、G2/M期细胞。结论:右旋一叶萩碱可以抑制黑色素瘤细胞的恶性增殖,其作用机制与诱导细胞凋亡、减少S期和G2/M期细胞有关。
Aim:To investigate the effect and mechanism of virosecurinine on proliferation and cell cycle of mouse melanoma cell line B16.Methods: B16 cells were treated with virosecurinine in vitro.Cell proliferation was measured by MTT assay.Cell cycle was detected by flow cytometry.Results: Virosecurinine inhibited proliferation of B16 in a time and dose-dependent manner with an IC50 value of 63.34 μmol/L at 72 h.Virosecurinine induced apoptosis on B16 cells and decreased cells of S and G2/M stages.Conclusion:Virosecurinine can inhibit malignant proliferation of melanoma cells.The mechanism is associated with induced apoptosis and decreased cells of S and G2/M stage in cell cycle.
