在橡胶树转录组测序的基础上,通过RT—PCR技术扩增得到了橡胶树的一个ADP核糖基化因子的鸟核苷酸交换因子基因(HbGNOM),该基因包含了一个4410bp的开放阅读框,编码了一个含1470个氨基酸的蛋白.生物信息学的分析结果表明:橡胶树HbGNOM蛋白与麻风树、木薯、蓖麻和拟南芥的GNOM蛋白高度相似,相似性分别为97%,96%,95%和84%.半定量分析的结果表明:当橡胶树受到白粉病菌侵染时,HbGNOM基因在叶片中的表达明显升高,但其对炭疽病菌的侵染不产生应答;HbGNOM基因在叶片中的表达可受植物激素乙烯(ET)的诱导,但其对水杨(SA)和茉莉酸(JA)的处理没有明显应答,因此,HbGNOM可能与乙烯信号传导途径有关,并且参与了橡胶树对白粉病的抗病反应.
The ARF guanine nucleotide exchange factors (ARF-GEF), together with small G protein, plays roles in vacuolar sorting and protein targeting. In the study, based on the transcriptome data of Hevea brasiliensis, an ARF-GEF was amplified by RT-PCR and named as HbGNOM. The ORF of HbGNOM was 4 410bp, encoding 1 470 amino acids. Bioinformatics analysis results showed that compared with that of GNOM from Jatropha curcas, Manihot esculenta, Ricinus communis and Arabidopsis thaliana, the homogenicity of amino acid sequence of Hb- GNOM is 97%, 96%, 95% and 84% ,respectively. Semi-quantitative RT-PCR analysis results indicated that the expression of HbGNOM in rubber tree leaves was significantly induced by ethylene ( ET), while there are no obvious responses to salicylic acid (SA) and jasmonic acid (JA). The expression of HbGNOM was significantly induced in rubber tree leaves inoculated with Odium heveae Steinm, but there is no responses observed when the rubber leaves were infected involve in the ET-mediated with Colletotrichum gloeosporoiodes. These findings suggested that HbGNOM might signaling pathway and the rubber tree resistance to powdery mildew.
