目的:探索核受体辅活化子(coactivator)PNRC(proline-rich Nuclear receptor coregulatory protein)在不同发育时期正常大鼠各种组织中的表达与其生理作用的关系;PNRC在人乳腺癌和相应癌旁组织中的表达与乳腺癌的关系.方法:以β-actin基因为参照,应用半定量RT-PCR和southern印迹技术,检测不同发育时期正常大鼠各种组织中PNRC的表达水平及其在人乳腺癌和癌旁组织中的表达.结果:PNRC在正常大鼠和胎鼠相应组织中广泛表达且无组织差异性(P>0.05);在成年大鼠各种组织中的表达明显高于胎鼠相应组织(P<0.01).PNRC在人乳腺癌中的表达水平明显低于相应癌旁组织(P<0.01).结论:PNRC是一种必需的、广泛表达的转录调节因子,它可能参与了机体多种正常组织的发育过程;在人乳腺癌和癌旁组织中的表达差异与它在乳腺癌发生中的潜在作用密切相关.
Objective: To study nuclear receptor coactivator PNRC (proline -rich nuclear receptor coregulatory protein) gene expression of various tissues in different developing period of rats and its potential role, and the difference in PNRC expression between human breast cancer and corresponding cancerous tissues and its relation to breast cancer. Methods: PNRC expression of various tissues in different developing period of rats, and the difference in expression levels between human breast cancer and corresponding cancerous tissues were examined by RT - PCR and southern blot. Results: PNRC was found to widely express in various tissues of grown rats and embryonic rats, and there was no difference between tissues in every kind of rat( P 〉 0.05 ). The expression level in various tissues of grown rats was higher than that of embryonic rats ( P 〈 0.01 ). The expression of PNRC in human non - cancerous breast tissue was obviously higher than that in corresponding cancer tissue( P 〈0.01 ). Conclusion: As an essential and conservative transcriptional regulator,PNRC may have a role in development of various normal tissues. The differ- ence of PNRC expression level between human breast cancer and corresponding non - cancerous tissues maybe closely associated with its role in progression of breast cancer.
