目的:研究赤芍总苷与黄芪多糖抑制血小板聚集的机制及其最佳配伍水平。方法:在对赤芍总苷与黄芪多糖分别采用不同剂量单味药实验的基础上,通过二次回归正交组合设计对赤芍总苷与黄芪多糖不同剂量配伍进行实验和分析。结果:赤芍总苷与黄芪多糖分别随着剂量的增加都能显著降低血小板的聚集率,赤芍总苷最佳剂量为125 mg.kg-1,黄芪多糖最佳剂量为300 mg.kg-1。赤芍总苷剂量和黄芪多糖剂量配伍与血小板聚集抑制率的关系模型为D=8.879+0.067 36 X1+0.164 75 X2+0.000 13 X1X2-0.000 542 X12-0.000 308 X22理论值与实际值能很好地吻合。结论:赤芍总苷与黄芪多糖对血小板聚集抑制有效,以88.908 mg.kg-1与284.3248 mg.kg-1水平配伍药效可能更好。
OBJECTIVE To investigate the mechanism of inhibition effect on platelet aggregation by total paeony glycoside(TPG)and Astragalus polysaccharide(APS),and to design the optimal compatible dosages of them.METHODS On the basis of using the different dosages of TPG and APS as the simple recipe respectively,quadratic regression orthogonal combination test was designed to study how the various dosages of TPG and APS produced together the synergistic effects.RESULTS Both TPG and APS can inhibit platelet aggregation remarkably and dose-dependently.The optimal dosages were 125 mg·kg-1 for TPG and 300 mg·kg-1 for APS.It was indicated that,the model for the relation between the inhibitory effect on platelet aggregation and the compatible dosages of TPG and APS was D=8.879+0.067 36X1+0.164 75X2+0.000 13X1X2-0.000 542X12-0.000 308X22.The theoretical value was equal with the practical one.CONCLUSION TPG concerted with APS in the inhibitory effect on platelet aggregation.
