目的探讨Nogo—A基因3'-侧翼区单核苷酸多态性(SNP)各等位基因及基因型在缺血性脑卒中(Ischemicstroke,IS)患者中的分布频率,并初步分析其基因型与Is的关系及其对血脂、脂蛋白水平的影响。方法采用单碱基延伸的PCR技术和DNA测序法检测202例Is患者及199例对照者的Nogo.A基因3I_侧翼区rs2588510C/T、rs887G/A多态性,分析各基因型及等位基因的分布频率;同时按常规方法测定血浆脂质、脂蛋白水平。结果Is病例组和正常组中Nogo-A基因3’.侧翼区rs2588510CC、CT和TT三种基因型的分布频率分别为19.3%和18.6%,48.5%和41.2%,32.2%和40.2%。rs887AA、GA和GG三种基因型的分布频率分别为64.9%和71.4%,31.7%和24.6%,3.5%和4.O%。Nogo—A基因3-则翼区rs2588510CFF、rs887G/A基因型频率和等位基因频率在IS组和对照组比较差异无显著性(P〉0.05)。结论Nogo-A基因3f-1则翼区rs2588510C/T、rs887G/A的多态性可能与1s的易感性无相关。
Objective To study the frequencies of allele and genotype distribution of neurite growth inhibitor-A (Nogo-A) gene 3'-flanking regions single nucleotide polymorphism (SNP), the relationship between Nogo-A gene 3'-flanking regions with ischemic stroke (IS) and analyze association the genotype of Nogo-A with IS and the effect ofNogo-A gene polymorphisms on plasma lipid and lipoprotein levels. Methods The Nogo-A gene 3'-flanking regions rs2588510 C/T and rs887 G/A polymorphism was examined by the polymerase chain reaction-single base extension (PCR-SBE) technique and DNA sequencing methods in 202 IS patients. Frequencies of allele and genotype of these Nogo-A gene polymorphism was analyzed compared with 199 normal persons. The plasma lipid and lipoprotein levels were measured by routine method. Results The frequencies of rs2588510 CC, CT and TT genotype were 19.3%, 48.5%, and 32.2% in IS patients respectively, while in controls were 18.6%, 41.2%, and 40.2%. The frequencies of rs887 AA, GA and GG genotype were 64.9%, 31.7% and 3.5% in IS patients respectively, while in controls were 71.4%, 24.6% and 4.0%. The Nogo-A gene 3'-flanking regions rs2588510 C/T and rs887 G/A polymorphism were not different between IS group and control group. ConcJusion The polymorphisms of Nogo-A gene 3'-flanking regions rs2588510 C/T and rs882 G/A probably are not associated with ischemic stroke.
