为了探讨温度对变异黄芪内生真菌多样性的影响。对采集于内蒙古腾格里变异黄芪的茎、叶和花,运用表面消毒法和尖端菌丝挑取法将组织块分别置于20℃、28℃和室温(变温)条件下进行内生真菌分离与纯化,采用形态学和ITS序列分析技术进行种属鉴定,并计算多样性指数。504块组织块中共分离到内生真菌410株,分属于10科15属23种,不同温度下分离到的内生真菌有所不同,Undifilum oxytropis为优势菌属,且在20℃时优势更为显著。多样性指数计算结果显示,香农-维纳指数(H)为1.85,辛普森指数(D)为0.269,均匀度指数(E)为0.59,不同温度下多样性指数不同,28℃培养香农-维纳指数最高,20℃培养辛普森指数最大。结果表明,温度对内生真菌分离率有一定影响,对Undifilum oxytropis分离率的影响极其显著,而对内生真菌的多样性影响也较为明显。
To ascertain the effect of different temperature on diversity of fungal endophyte from Astragalus variabilis,and Astragalus variabilis samples in Tengeli County of Inner Mongolia were collected. The method of surface disinfection was used to isolate and purify the fungal endophyte in stems,leaves and flowers. And then morphology techniques and ITS sequence analysis were used to identify the species,and the diversity index was calculated. The results showed that 410 fungal endophyte were isolated from 504 samples,which included 23 kinds of fungal endophytes. There were different kinds of fungal endophyte under different temperature,and Undifilum oxytropis was the dominant strain,especially at 20 ℃. The Shannon- Weiner index was 1. 85,Simpson diversity index was0. 269,and evenness index was 0. 59. The diversity index was different in different temperature. The Shannon- Weiner index was biggest in 28 ℃,and simpson diversity index was the biggest in 20 ℃. The results show that the influence of temperature to isolation rate of fungal endophyte is not significant,but the isolation rate of Undifilum oxytropis and diversity of fungal endophyte is significant.
