目的:建立远志糖蛋白的最佳提取工艺。方法:以蛋白得率为评价指标,从p H值、温度、盐浓度、液料比、提取时间5个方面进行单因素试验,根据单因素试验结果,选取其中影响较大的三个因素,运用响应曲面方法考察远志糖蛋白的最佳提取工艺,并对最佳提取工艺进行工艺验证。结果:采用p H=7.85的Tris-HCl(Na Cl浓度0.1mol/L)缓冲液,以液料比8m L/g、温度33℃水浴提取1.5h,得到的糖蛋白得率最高为5.96%。结论:采用响应曲面方法建立的模型预测相对准确,可以较好地优选出远志糖蛋白的最佳提取工艺。
Objective: To establish the optimal extraction technology of Polygala tenuifolia Willd. glycoprotein. Methods: With protein yield as index, from the p H, temperature, salt concentration, liquid-solid ratio, extraction time five aspects of single factor experiment, according to the results of single factor test, select three factors which affect larger in these five aspects, by response surface method to study the optimum extraction process of Polygala tenuifolia Willd. glycoprotein, and verification of optimum extraction technology. Results: In the conditions of the p H=7.85 Tris-HCl(Na Cl concentration of 0.1mol/L) buffer, liquid to material ratio 8m L/g, temperature of 33℃ water bath extraction for 1.5h, the glycoprotein is highest rate for 5.96%. Conclusion: The established model which adopts response surface method predicts exactly, and could well optimize the extraction process of Polygala tenuifolia Willd. glycoprotein.
