采用ISSR分子标记技术,对斜带髭鲷3个群体(柘林湾人工繁育群体、厦门湾人工繁育群体和厦门近海捕捞野生群体)的遗传多样性水平和群体遗传结构进行了研究.筛选出17个ISSR引物对3个群体共54个样品进行扩增,共得到145个清晰的扩增位点,其中多态性位点80个,多态位点百分率为55.17%.Popgene分析结果表明:厦门野生群体的遗传多样性水平最高(PPB=51.72%,h=0.1826,I=0.2719),略高于其他2个人工繁育群体的遗传多样性水平.群体间的Nei’s遗传分化系数和基因流分别为:0.0876和5.2098.表明斜带髭鲷3个群体的遗传分化尚未达到种群的分化水平.本文通过对斜带髭鲷野生群体及人工繁育群体遗传结构和遗传多样性水平的研究,为其种质资源的科学管理提供理论依据.
The genetic diversity and population genetic structure of 54 H. nitens samples from three stocks (18 from cultured stock in Zhelin Bay, 18 from cultured stock in Xiamen Bay and 18 from wild stock in Xiamen offing) were studied using the inter-simple sequence repeat (ISSR) marker. The ISSR bands can be differentiated highly in 17 primers after screening. A total of 145 discernible DNA fragments were observed, among which 80 fragments (55.17%) being polymorphic. The percentage of polymorphic (PPB), Nei's gene diversity and Shannon's index (calculated in POPGENE(in small capital)) showed that the wild stock in Xiamen had the highest genetic variation (PPB:51.72%, h=0. 182 6, I=0. 271 9 ). The inter-stocks Nei's gene diversity coefficient and gene flow were 0. 0876 and 5. 2098 respectively, indicating that the genetic difference among three stocks hasn't reached the population level yet.
