目的从mRNA水平观察三七总皂苷对肝纤维化大鼠Ⅰ型前胶原及TGF-β1的影响。方法用四氯化碳复合因素诱导大鼠肝纤维化模型。同时给予三七总皂苷治疗,秋水仙碱治疗组作对照,病理HE染色观察纤维化程度分期,用RT-PCR进行Ⅰ型前胶原mRNA和TGF-β1mRNA的半定量检测。结果三七总皂苷能抑制肝纤维化大鼠Ⅰ型前胶原及TGF-β1的合成表达,减轻肝纤维化程度。结论三七总皂苷具有抗肝纤维化作用,干预大鼠肝纤维化可能与减少TGF-β1含量有关。
Objective To observe the effect of total saponins of panax notoginseng(SPN) on the production of collagen I and TGF-β1 in experimental hepatic fibrosis rats induced by carbon tetracloride.Methods Experimental hepatic fibrosis model was made by intracutaneous injection of CCl4 twice a week in rats for 12 times.From the first day after the CCl4 injection,SPN(60 or 30 mg/kg) were given intracutaneously once a day for 42 days.The rats were sacrificed at the end of the 43rd days.The expression of type collagen I and TGF-β1 mRNA of hepatic tissue were measured by RT-PCR method.Results The expression of type collagen I and TGF-β1 mRNA were more in liver hepatic fibrosis model group than those in SPN treatment group.Conclusion SPN inhibited the expression of the type collagen I and TGF-β1 and could slow down the progression of hepatic fibrosis.
