本试验旨在研究谷氨酰胺对饲喂高精料奶山羊盲肠黏膜形态及炎症因子mRNA表达的影响。采用随机区组设计,将12头健康奶山羊随机分为对照组( n=6,饲喂高精料)和谷氨酰胺组( n=6,谷氨酰胺添加量为50 g/d)。试验期28 d。每组各选取4头山羊进行屠宰,取盲肠黏膜,观察组织形态学变化,检测盲肠黏膜炎症因子mRNA的表达量、盲肠内容物挥发性脂肪酸浓度。结果表明:添加谷氨酰胺对盲肠食糜中挥发性脂肪酸、乳酸及内毒素浓度无显著影响(P>0.05);添加谷氨酰胺保持了盲肠黏膜的完整性,紧密连接得到增强;添加谷氨酰胺显著降低了盲肠黏膜肿瘤坏死因子αmRNA表达量( P<0.05),但对干扰素γ、白细胞介素1β、白细胞介素6和白细胞介素10 mRNA表达量无显著影响( P>0.05)。结果提示,高精料条件下,添加谷氨酰胺可加强盲肠黏膜紧密连接,一定程度上缓解盲肠黏膜的炎症反应,有助于维持后肠黏膜屏障的正常功能。
The objective of the present study was to investigate the effects of glutamine ( Gln) on cecal mucosa morphology and inflammatory cytokine mRNA expressions of dairy goats fed a high concentrate diet.Twelve healthy dairy goats were randomly assigned to control group ( n =6, fed a high concentrate diet) and Gln group ( n=6 , supplemental level of Gln was 50 g/d) .After a 28-day experimental period, four goats in each group were slaughtered to collect cecal mucosa.The changes of tissue morphology of cecal mucosa, the mR-NA expression levels of inflammatory cytokines in cecal mucosa and the concentrations of volatile fatty acids in cecal contents were measured.The results showed as follows: Gln supplementation did not affect the concen-trations of volatile fatty acids, lactic acid and endotoxin contents in cecal (P〉0.05); the supplementation of Gln played roles in keeping the integrity and enhancing tight junction of cecal mucosa; the supplementation of Gln significantly decreased the mRNA expression level of tumor necrosis factorα( P〈0 .05 ) , but had no sig-nificant effects on that of interferon γ, interleukin 1β, interleukin 6 and interleukin 10 in cecal mucosa ( P〉0 .05 ) .It is suggested that Gln supplementation can enhance cecal mucosa tight junctions release cecal mucosal inflammatory response, to a certain extent, which is benefit for maintenance of mucosal barrier function of hindgut under the condition of the high concentrate diet.
