观察高碘诱发自身免疫性甲状腺炎动物模型中CD4^+ CD25^+调节性T细胞、Th17细胞的变化及溴氯哌喹酮(HF)干预对其影响选取NOD.H-2h4雌鼠饮0.005%碘化钠水,皮下注射HF干预.观察淋巴细胞浸润情况并进行甲状腺炎症程度评分,测定甲状腺球蛋白抗体(TgAb)水平,分析CD4^+ CD25^+ Foxp3^+调节性T细胞、Th17比例的变化,实时定量PCR检测Foxp3、IL-17和RORγt mRNA表达水平.NOD.H-2^h4小鼠饮高碘水后甲状腺炎的发生率明显高于对照组,甲状腺组织有不同程度的淋巴细胞浸润,甲状腺相对重量及TgAb水平均较对照组明显升高(P<0.05).发生自发性自身免疫性甲状腺炎(SAT)的小鼠脾细胞中CD4^+ CD25^+ Foxp3^+调节性T细胞所占比例和Foxp3 mRNA表达量均较对照组明显降低(P<0.01);Th17所占比例和IL-17、RORγt mRNA表达水平均较对照组明显升高(P<0.05).与SAT非HF治疗组相比,HF治疗组CD4^+ CD25^+ Foxp3^+调节性T细胞的比例和Foxp3 mRNA表达量均无明显变化(P>O.05);但Th17比例及IL-17、RORγt mRNA表达水平均明显减低(P<0.01).同时,HF治疗组TgAb滴度以及淋巴细胞浸润程度均较同周龄SAT非HF治疗组明显降低(P<0.05).
The changes of CD4^+ CD25^+regulatory T cells and Thl7 cells in NOD.H-2h4 mice with iodineinduced autoimmune thyroiditis were investigated.NOD.H-2^h4 mice were fed with 0.005 % NaI (50 mg/L NaI) water for 8 weeks to establish the animal models of autoimmune thyroiditis (AIT).At the same time,the mice were fed with 0.005% Nal water supplied with halofuginone (HF) for8 weeks.Thyroid tissues were removed and the relative thyroid weights were measured.The severities of lymphocytic infiltration in thyroids were observed.Compared with control animals,SAT mice without HF treatment had lower numbers of CD4^+CD25 + Foxp3^+T cells in the splenic mononuclear cells and reduced expressions of Foxp3 mRNA in splenocytes (P〈0.01) ; higher numbers of Th17 cells in the splenic mononuclear cells and increased expressions of IL-17 and RORγ tmRNA in splenocytes (P〈0.05) ; relative thyroid weight and serum thyroglobulin antibody(TgAb) titers increased obviously (P〈0.05),lymphocytic infiltration could be found in thyroids with scores from 1 + to 4+.After HF treatment for 8 weeks,compared to SAT mice without HF treatment,the percentage of CD4 + CD25 + Foxp3 + T cells and expressions of Foxp3 mRNA revealed no change in SAT/ HF treated group (P〉0.05) ; the percentage of Th17 cells and expressions of IL-17 and RORγt mRNA reduced significantly in SAT/HF treated group (P〈0.01).Simultaneously,the relative thyroid weight and serum TgAb titers decreased significantly (P 〈 0.01),histomorphologic appearances of thyroids improved with lower scores of thyroiditis.
