利用离子对色谱-电化学检测器建立一种高灵敏、快速的检测方法,用于脑组织中硫醇和鸟苷代谢物的同时测定.采用Agilent SB-AQ色谱柱,以100 mmol/L V(磷酸缓冲盐,pH 2.0,含350 mg/L辛烷基磺酸钠):V(甲醇)=95:5为流动相进行分离,检测电压为800 mV.所测的6种硫醇和鸟嘌呤类物质在15 min内得到完全分离,且在测定浓度范围内线性良好.检出限为22.6~56.9 nmol/L,不同加标水平的回收率为94.4%~106.3%,变异系数为0.21%~3.85%.该方法可用于脑组织中硫醇和鸟嘌呤类物质的同时测定.
A highly sensitive,rapid detection method was established for the determination of thiols(glutathione and cysteine) and guanosine metabolites(guanosine,guanine,xanthine and uric acid) in the rat brain using high performance liquid chromatography with an electrochemical detector.100 mmol/L phosphate buffer(pH 2.0,containing 350 mg/L octane sulfonic acid sodium salt,95%)-methanol(5%) was used as the mobile phase and the detection potential was set to 800 mV.All of the six compounds were of good linearity in three orders of magnitude,and the detection limit was 22.6-56.9 nmol/L.The recovery of different spiked levels ranged from 94.4%to 106.3%and the coefficient of variations was from 0.21%to 3.85%.The constructed method can be employed for the determination of the six compounds in the rat brain.
