目的 探讨原因不明复发性流产(URSA)患者蜕膜组织中Th17/Treg免疫平衡情况.方法 应用流式细胞技术检测URSA患者和正常早孕者蜕膜组织中Th17和Treg细胞频率,应用real-time PCR方法检测转录因子RORc和Foxp3 mRNA水平.结果 URSA患者蜕膜组织中Th17细胞在淋巴细胞和CD4+T细胞中的频率显著高于正常早孕者(P〈0.001),URSA患者蜕膜组织中Treg细胞在淋巴细胞和CD4+T细胞中的频率显著低于正常早孕者(P〈0.001).URSA患者蜕膜组织中RORc mRNA水平明显高于正常早孕者,而Foxp3 mRNA水平显著低于正常早孕者(P〈0.001).结论 URSA患者蜕膜组织中存在明显的Th17/Treg免疫失衡,可能是URSA发生的重要原因.
Objective To investigate the imbalance Thl7/Treg in deciduas of unexplained recurrent spontaneous abortion (URSA). Methods In URSA patients and health early pregnant subjects, Thl7 and Treg cells frequencies were measured by flow cytometry; while RORc and Foxp3 mRNA levels were measured by real - time PCR. Results There was significant increase in decidual Thl7 cell frequency and RORc mRNA level in URSA patients (P 〈 0. 001 ). Meanwhile, there was significant reduction in decidual Treg cell frequency and Foxp3 mRNA level in URSA patients ( P 〈 0. 001 ). Conclusion The decidual Thl7/Treg imbalance plays an important role in URSA.
