目的探讨桑白皮Mori Cortex有效部位对心衰大鼠心功能及肾素-血管紧张素-醛固酮系统(RAAS)的影响。方法多次腹腔注射2 mg/m L阿霉素诱导SD大鼠慢性心衰。将成模大鼠随机分为模型组,羧甲基纤维素钠(CMCNa)组,卡托普利组,桑白皮脂肪油、醇沉、30%乙醇洗脱、50%乙醇洗脱、80%乙醇洗脱、水煎液组,每组8只,灌胃给药4周;另取8只SD大鼠作为空白组,灌胃蒸馏水4周。采用大鼠代谢笼法收集6 h尿液;运用超声心动图检测大鼠左室舒张末内径(LVEDD)和收缩末内径(LVESD),并计算左室短轴缩短率(LVFS)及射血分数(LVEF);光镜观察大鼠心肌组织形态变化;酶联免疫吸附试验(ELISA)检测大鼠血液中脑钠肽(BNP)、心肌钙蛋白I(c Tn I)、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)、肿瘤坏死因子-α(TNF-α)的水平。结果桑白皮30%乙醇洗脱组分能改善心衰大鼠心功能(P〈0.01),降低BNP、c Tn I、TNF-α水平(P〈0.01);同时增加大鼠尿量,降低渗透压(P〈0.01),且能够降低AngⅡ、ALD水平(P〈0.01)。结论桑白皮30%乙醇洗脱组分具有更好的抗心衰作用。
AIM To explore the effective components of Mori Cortex that contribute to cardioprotective and renin-angiotensin-aldosterone system( RAAS). METHODS Chronic heart failure( CHF) model of male SD rats were established by intraperitoneal injection of Doxorubicin. The experimental rats were randomly divided into model group,CMC-Na group,Catopril group,fatty oil,ethanol precipitation,30%,50%,80% ethanolic fraction and decoction of Mori Cortex groups( n = 8),and the gastric gavage was fed for four weeks. Another eight SD rats were taken as control group,and were gavaged with distilled water for four weeks. The impact on 6 h urine volume of rats was observed by the metabolic cage method. The left ventricular end diastolic diameter( LVEDD),left ventricular end systolic diameter( LVESD) were detected by echocardiography and left ventricular fractional shortening( LVFS) and ejection fraction( LVEF) were calculated. Heart tissues were visualized under light microscope. The levels of brain natriuretic peptide( BNP),c Tn I,Ang Ⅱ,ALD and TNF-α in blood were measured by ELISA.RESULTS The 30% ethanol fraction of Mori Cortex could obviously improve the cardiac function( P〈0. 01)and down-regulate the levels of BNP,c Tn I and TNF-α( P〈0. 01). It also increased urine of the rat,reduced osmotic pressure( P〈0. 01). It could obviously down-regulated the levels of Ang Ⅱ and ALD( P〈0. 01).CONCLUSION The 30% ethanol fraction of Mori Cortex has much better cardiotonic effect.
