目的:明确Cosmc基因在卵巢癌细胞中的生物学功能。方法:本研究采用慢病毒转染技术,在卵巢癌细胞A2780和SKOV3中过表达Cosmc基因,MTT实验、细胞凋亡实验以及Transwell侵袭实验对过表达Cosmc后卵巢癌细胞在生长、凋亡、侵袭等方面的影响。结果:慢病毒转染卵巢癌细胞A2780和SKOV-3后,Cosmc的蛋白表达显著提高;MTT结果显示,与空载体对照组相比,过表达Cosmc的卵巢癌细胞生长能力显著减弱;流式细胞术结果表明Cosmc过表达的卵巢癌细胞凋亡数较空载体对照组细胞显著增加;Transwell实验显示Cosmc过表达的细胞侵袭和迁移能力较空载体对照组显著减少。结论:卵巢癌细胞系中过表达Cosmc基因能抑制卵巢癌细胞的生长和侵袭并促进细胞凋亡。
Objective: To determine the biological function of Cosmc gene in ovarian cancer cells. Methods: Lentivirus technology was used to build Cosmc over expression vector in ovarian cancer cells SKOV3 and A2780.MTT assay, apoptosis experiments, and Transwell assay were used to test cell growth, cell apoptosis, cell invasion by over expression of Cosmc. Results: The expression of Cosmc was greatly improved by Lentiviral transfection in ovarian cancer cells A2780 and SKOV-3; MTT results showed that compared with the empty vector, the ability to grow was significantly weakened in ovarian cancer cells which Cosmc was over expressioned; Flow cytometry results showed apoptosis was significantly increased compared with the control; Transwell experiment showed that compared with the empty vector, the ability of invasion and metastasis was significantly weakened in ovarian cancer cells which Cosmc was over expressioned. Conclusion: The over expression of Cosmc in ovarian cancer cell lines can inhibit the growth and invasiveness and promote apoptosis.
