通过建立兔动脉粥样硬化模型,收集各时期外周血相关血脂数据,确定动物模型的建立情况,并结合解剖后得到的组织切片进一步确定其病理发展。然后用实时荧光定量PCR检测单核细胞整联蛋白beta2的mRNA表达水平,激光共聚焦和流式细胞术检测beta2的蛋白表达水平。结果表明,整联蛋白beta2的mRNA表达水平和蛋白表达水平随建模时期推移均有显著升高。提示整联蛋白beta2的表达量随动脉粥样硬化的发生发展不断升高,可能对临床诊断由动脉粥样硬化引起的心肌梗塞有一定的预测作用。
The model of rabbits atherosclerosis was ensured by collecting the peripheral blood lipids data and combining with the tissue sections to further determine the pathological development.The expression level of mRNA and protein were detected by real-time fluorescence quantitative PCR and laser scanning confocal microscopy and flow cytometry respectively.The expression level of mRNA and protein of the whole protein beta2 increased significantly with the passage of time.It suggests that the expressional volume of the whole protein beta2 continuously increased with atherosclerosis development.It may be a predictive way for the clinical diagnosis of myocardial infarction caused by atherosclerosis.
