目的:观察“劳倦过度,房室不节”诱发的肾阳虚模型小鼠舌象变化。方法:选健康雄性昆明种小白鼠45只,随机分为对照组、模型组和治疗组3组,各15只。用强迫小鼠游泳法造成劳倦过度;以Colldege效应诱导雄性小鼠房室不节,建立肾阳虚小鼠模型。用金匮肾气丸作为治疗药物,6周时,观察各组小鼠舌象,做石蜡切片,HE染色,显微镜观察拍片。结果:模型组呈淡白舌,齿痕明显;治疗组基本为淡白舌,齿痕不明显。模型组舌乳头密度、舌分裂相频数较对照组明显下降(P〈0.01),上皮层厚度、角质层厚度较对照组显著增加(P〈0.05);治疗组较模型组乳头密度显著增加,上皮层厚度显著减小(P〈0.05),舌分裂相频数增加,角质层厚度下降,但无统计学意义。结论:肾阳虚模型小鼠舌黏膜病理改变以上皮增殖减慢、代谢降低、萎缩和退变为主要特征。
Objective: To observe the changes of the tongue condition of mouse models with kidney-Yang deficiency caused by excessive lassitude and sexual activity. Methods: 45 healthy male Kunming mice were randomly divided into 3 groups with 15 mice in each group. The control groups are the kidney-Yang deficiency model group and the treatment group. The model group is prepared with compulsive swimming and excessive sexual activity induced by Colldege effect everyday for 6 weeks. The treatment group was treated the same way as the model group, but was fed with Jingui Shenqi pill additionally. All tongue conditions were observed under microscope after HE staining. Results: The model group animals had the obvious crenated tongues in comparison with the treatment group. Comparing with the control group, the model group had the mantel changes as follows: the papillae of the lamina propria became less in density, the nucleus division phase frequency of basal layer cells decreased(P〈0.01), the thickness of the lamina epithelium and corneum in-creased obviously (P 〈 0.05). Comparing with the model group, the density of the papillae increased greatly( P〈 0.05) and the thickness of the lamina epithelium greatly decreased in the treatment group. The nucleus division phase frequency of basal layer cells tended to increase and the thickness of the corneum tended to decrease, but there is no sense statistically. Conclusion: Experimental study shows the tongue pathologic changes in this lddney-Yang deficiency group were characterized by decreased opitheliosio and metabolism and atrophic and retrograde degeneration.
