牛磺酸对高糖刺激Mller细胞VEGF和TauT表达的影响

目的通过检测高糖和牛磺酸培养对大鼠视网膜Muller细胞中血管内皮细胞生长因子(vascularen—dothelialgrowthfactor,VEGF)和牛磺酸转运蛋白(taurinetransporter,TauT)表达的影响,探讨牛磺酸保护糖尿病引起视网膜损伤的机制。方法实验分6组,分别是正常对照组、牛磺酸对照组、高糖模型组、低、中、高剂量牛磺酸处理高糖组。用免疫细胞荧光双标鉴定Muller细胞,RT-PCR和免疫细胞荧光双标检测Muller细胞中VEGF和TauT的表达。结果体外25mmol/L高糖培养使Muller细胞VEGFmRNA及蛋白表达增加。10mmol/L牛磺酸可有效抑制高糖诱导的VEGF上调表达(P〈O.05);高糖诱导Muller细胞中TauT表达抑制,引起TauTmR—NA及蛋白表达明显下降。牛磺酸干预可上调Muller细胞中TauT的表达(P〈0.05),提高细胞内外牛磺酸的转运能力。结论体外高糖培养诱导Muller细胞VEGF表达增加,TauT表达下降。牛磺酸通过降低VEGF表达、提高牛磺酸转运能力达到保护视网膜的作用。该实验结果可为牛磺酸用于糖尿病早期视网膜病变的防治提供重要的实验依据。

英文摘要:

Objective To investigate the effect of taurine on expression of vascular endothelial growth factor (VEGF) and taurine transporter (TauT) in rats retinal Muller cells cultured with high glucose and observe the protection mechanism of taurine against diabetic retinopathy. Methods The cells was divided into normal control group (Control), taurine control group (Tau), high glucose group (HG) and high glucose plus different dosage taurine group (HG+L-Tau, HG+M-Tau and HG+ H-Tau). The morphologia were identified by confocal microscopy. The changes of VEGF and TauT expressed in Muller cells were studied by immunofluorescence staining method in a qualitative way and RT-PCR in a quantitative way. Results High glucose could significantly increase the expression of VEGF in Muller cells (P〈0. 05). 10 mmol/L taurine intervention could significantly inhibite the up-regulation of VEGF in Muller cells induced by high glucose (P〈0. 05). Our study also suggested that the expression of TauT reduced in high glucose group and could be stimulated by taurine supplementation (P〈0. 05). Conclusion Our studies reveal that taurine ameliorates diabetic retinopathy possibly via decreasing the expression of VEGF in Muller cells, increasing the ability of taurine transporter of Muller cells, which suggest a possible utility of taurine as an adjuvant therapeutic agent in the management of diabetes and its complications.
