以金刚酸丙酯基碲(ADA-Te-ADA)作为疏水性客体分子,CH3-β-环糊精作为亲水性主体分子,基于超分子主客体自组装作用构建了一种水溶性谷胱甘肽过氧化物酶模拟物(CD-Te-CD),并研究了其自组装的结构和催化机理。利用动态光散射(DLS)、扫描电镜(SEM)和透射电镜(TEM)等阐明了仿生酶CD-Te-CD2.2在水溶液中可以形成囊泡结构。酶的催化活力测定表明,CD-Te-CD2.2囊泡结构中的疏水空腔可以模拟天然谷胱甘肽过氧化物酶(GPx)的识别位点,有利于提高其对疏水性底物的识别能力。动力学研究表明,CD-TmCD2.2具有典型的酶学催化行为。本研究不仅建立了一种水溶性GPx模拟酶的高效制备方法,而且还为构建其他新型的超分子自组装仿生硒酶船棋依据,

英文摘要:

Based on the supramolecular host-guest self-assemble, a soluble glutathione peroxidase mimic with the optimum structure (CD-Te-CD2.2 ) was constructed. Herein, 3,3'-tellurobis (propane-3,1-diyl) diadamantanecar- boxylate (denoted as ADA-Te-ADA) was employed as hydrophobic guest molecule. CH3-β-cyclodextrin was used as hydrophilic host molecule. The self-assembled structure and catalytic mechanism of CD-Te-CD2. 2 were detailed investi- gated. DLS (Dynamic Light Scattering), SEM and TEM suggested that CD-Te-CD2 .2 could form the vesicle-like aggregates with hollow structure in water. And saturation kinetics measurement indicated that CD-Te-CD2.2 exhibited the typical enzyme catalytic behavior. Furthermore, benefited from the hydrophobic microenvironment in vesicle-like aggregates, CD-Te-CD2. 2 exhibited the specific recognition ability for hydrophobic substrates, which was one of the vi tal characteristics of native glutathione peroxidase (GPx). The successful construction of CD-Te-CD2. e not only provides an efficient preparation method for soluble GPx mimic but also highlights for the construction of other giant su pramolecular biomimetic enzymes.
