目的:探讨小鼠耳蜗螺旋神经元上是否存在五羟色胺(serotonin,5HT)1A受体。方法:采用出生后3天~6天的乳小鼠耳蜗进行冰冻切片,运用免疫荧光技术检测耳蜗螺旋神经元上五羟色胺1A受体的表达。结果:实验组乳小鼠耳蜗螺旋神经元胞浆内显示阳性表达信号,阴性对照组未发现阳性信号。结论:五羟色胺1A受体应存在于小鼠耳蜗螺旋神经元上,螺旋神经元兴奋时,五羟色胺作为神经递质与螺旋神经元上的五羟色胺1A受体结合,对其兴奋性进行调节。
Objective: Locate the expression of serotonin receptor 1A in the spiral ganglion. Methods: In the cochlear frozen slides of postnatal mice which were bom between three to six days, the localization of the serotonin receptor 1A in the spiral ganglion was labeled with immunofluorescenee technique. Results: Positive signals were seen in the cytoplasm of spiral ganglion. As a negative control without serotonin receptor 1A antibody, no signal was identified in the cytoplasm of spiral ganglion. Conclusion: Serotonin receptor 1A is located in the spiral ganglion. It indicates that serotonin as a neurotransmitter maybe combine with the serotonin receptor 1A to play a role in modMating the auditory transmission of the spiral ganglion when it is stimulated by the hair cells to be excitatory.
