γ-氨基丁酸(gamma-aminobutyric acid,GABA)受体是昆虫体内重要的抑制性神经递质受体,是多种杀虫剂的重要作用靶标,其结构与相关农药的毒理性质和害虫的抗药性密切相关。对昆虫GABA受体进行分离纯化并研究其立体结构,对于研究开发高效、低毒、安全的新农药及克服害虫抗药性均具有重要的指导意义。GABA受体的分离、纯化方法与其分子质量大小、带电性和特异性亲和力有关。对GABA受体的主要分离纯化方法:沉淀法、密度梯度离心法、离子交换色谱法、羟基磷灰石色谱法、凝胶过滤法和亲和色谱法等进行了综述,评述了不同纯化方法的优缺点,并总结了分离所用各种去污剂对该受体提取效率的影响。
As an important inhibitory neurotransmitter receptor in the insects,gamma-aminobutyric acid (GABA) receptor acts as a concerned target for many insecticides. The structure of GABA receptor is closely related to the insecticide resistance and toxicology. Therefore, isolation/purification of the GABA receptor of insect and further study on the spatial structure will shed light on the development of high-efficient,low toxic and safe insecticide as well as overcoming the pesticide resistance. Various isolation/purification methods have been established according to the molecular weight, charge and specific affinity of GABA receptor. The methods of GABA receptor isolation/purification including precipitation, density gradient centrifugation, ion exchange chromatography, hydroxyapatite chromatography, gel filtration, and affinity chromatography, etc, were reviewed. The advantages and disadvantages of these methods were discussed, and the influence of the detergents on the extraction efficiency of GABA receptor were summarized.
