目的 探讨温肾壮骨方(补骨脂,蛇床子,制附子)对乳腺癌骨转移Tac1/NK1R通路相关蛋白表达的影响.方法 左心室注射肿瘤细胞建立乳腺癌骨转移裸鼠模型(n=50),随机分为温肾壮骨方高、中、低剂量组、模型对照组和阳性对照组(每组n=10),连续给药6周.组织病理学观察骨转移组织的形态学变化;免疫组化法检测骨转移灶中Tac1/NK1R通路相关蛋白的表达.结果 与模型对照组相比,温肾壮骨方组骨组织损伤明显缓解,骨转移灶中的Tac-1、NK1R、SDF-1α、CXCR4的蛋白表达明显降低(P<0.05或P<0.01).结论 温肾壮骨方对乳腺癌骨转移裸鼠模型具有一定的治疗作用,其机制可能与下调Tac1/NK1R通路相关蛋白表达有关.
AIM To investigate the effects of Wenshen Zhuanggu (WSZG) (Psoraleae Fructus, Cnidii Fructus, Aconiti lateralis Radix praeparata, Aconiti lateralis Radix praeparata ) recipe on Tacl/NKIR signaling pathway pro- teins in nude mice with bone metastatic of breast cancer. METHODS A nude mouse model for breast cancer bone metastasis was established via peritoneal injection of MDA-MB-231BO cells. Fifty mice were divided into five groups: model control group, positive control group, and three dosages of WSZG recipe groups (high, middle and low dose), and all drugs were administered orally for six weeks. Pathology of bone metastatic tissues was observed under a light microscope, and the related protein expressions in Tacl/NK1R signaling pathway were determined by immunohisto- chemical technique. RESULTS Compared with model mice, WSZG recipe significantly alleviated bone injury symptom and simultaneously reduced Tac-1, NK1R, SDF-lα, CXCR4 protein expressions in bone metastatic tissues (P 〈 0. 05 and P 〈 0. 01 ). CONCLUSION WSZG recipe could inhibit breast cancer bone metastasis in nude mice and the mechanism might be associated with down-regulation of proteins in Tacl/NK1R signaling pathway.
